Engeland, 19de eeuw. Dr. Dolittle, een bekend geleerde, krijgt het steeds moeilijker om met zijn patiënten te communiceren. Het is net alsof hij gemakkelijker met de dieren kan praten. Polynesia, zijn papegaai, geeft hem de tip om taallessen van de dieren aan te leren, en zo heeft hij wel 500 verschillende dierendialecten geleerd. Dolittle wordt een graag geziene gast bij de dieren en aarzelt dan ook niet om een krokodil van haar tandpijn af te helpen. Maar de dokter heeft ook vijanden: generaal Bellowes, kantonrechter en berucht vossenjager!
